Как следует из описание ошибки, файл VBoxManage не обнаружился в переменной $PATH.
Необходимо любым удобным способом обнаружить путь к файлу отвечающему за запуск конфигурации Vbox в вашем дистрибутиве и после этого добавить путь к нему в переменную:
export PATH=/some/path

Сразу замечу, что актуальный рабочий способ сборки предложенной в курсе, это сборка под управлением ОС Windows, на котором установлен Virtual Box, Packer скачан с сайта. Операции по запуску сборки можно выполнять в Powershell.
